اثرات استفاده از گندم عملآوری شده به روشهای مختلف بر عملکرد رشد و قابلیت هضم مواد مغذی در برههای پرواری
سابقه و هدف: گندم یکی از محصولات غالب کشت در ایران میباشد که میتواند در تغذیه دام مورد استفاده قرار گیرد. استفاده از اوره به عنوان منبع ازت و در ترکیب با هیدروکسیدسدیم و فرمالدئید میتواند به صورت آهسته رهش مورد استفاده میکروارگانیسمها قرار گیرد. از طرفی فرآوریهای مختلف فیزیکی میتواند بر تجزیهپذیری مواد مغذی آن در شکمبه و همچنین قابلیت هضم آنها در قسمتهای بعدی شکمبه اثر گذار باشد. بنابراین هدف از این مطالعه بررسی اثرات فرآوریهای مختلف گندم بر ترکیبات شیمیایی، عملکرد رشد و قابلیت هضم در تغذیه برههای پرواری میباشد. مواد و روشها: تعداد 30 راس بره نر نژاد افشاری با میانگین وزن اولیه 3±21/31 کیلوگرم مورد استفاده قرار گرفت. این آزمایش در قالب طرح کاملاً تصادفی با 6 تیمار شامل: 1- جیره شاهد: گندم کامل، 2-گندم آسیاب شده، 3-گندم عملآوری شده با اوره و هیدروکسیدسدیم و آسیاب شده، 4-گندم عملآوری شده با اوره و فرمالدئید و آسیاب شده ، 5-گندم عملآوری شده با اوره و هیدروکسیدسدیم و پلت شده و 6-گندم عملآوری شده با اوره و فرمالدئید و پلت شده انجام گرفت. به هر تیمار 5 راس بره پرواری تعلق گرفت. 14 روز عادت پذیری و 84 روز دوره آزمایشی برای دامها در نظر گرفته شد. از روز 80 از جیرههای آزمایشی و مدفوع نمونهگیری شد و قابلیت هضم ظاهری مواد مغذی با روش خاکستر نامحلول در اسید اندازهگیری شد. یافتهها: نتایج آزمایش نشان میدهد که عملآوریهای فیزیکی-شیمیایی گندم به شکل معنیداری منجر به افزایش وزن روزانه بیشتر نسبت به تیمار بدون عملآوری شد (05/0>P). همچنین تیمار پنجم (اوره+هیدروکسیدسدیم+پلت شده) به شکل معنیداری نسبت به تیمارهای اول (بدون عملآوری) و تیمار دوم (آسیاب شده) افزایش وزن روزانه بالاتری نشان داد (05/0>P). از سویی نتایج نشان داد که عملآوری فیزیکی-شیمیایی منجر به وزنگیری بالاتری در انتهای دوره نسبت به تیمار بدون عملآوری و تیمار به تنهایی آسیاب شده، میشود (05/0>P). نتایج حاصل از عملآوریهای فیزیکی-شیمیایی گندم حاکی از عدم تاثیر معنیدار بر روی متوسط مصرف خوراک روزانه میباشد ( 05/0<P). نتایج در رابطه با ضریب تبدیل غذایی مشابه با افزایش وزن روزانه میباشد و نشان میدهد که فرآوریهای فیزیکی-شیمیایی منجر به بهبود ضریب تبدیل غذایی نسبت به تیمار بدون عملآوری شد (05/0>P) و در پایان دورهی پروار تیمارهای پنجم (اوره+هیدروکسیدسدیم+پلت شده) و ششم(اوره + فرمالدئید + پلت شده) کمترین ضریب تبدیل غذایی را به خود اختصاص دادند. عملآوری های فیزیکی-شیمیایی به شکل معنیداری سبب بهبود قابلیت هضم ماده خشک، فیبر نامحلول در شویندهی خنثی، فیبر نامحلول در شویندهی اسیدی و نشاسته دانه گندم شد (05/0>P) در عین حال تاثیر معنیداری بر قابلیت هضم ماده آلی و پروتئین خام این دانه غله نداشت (05/0<P). نتیجهگیری: نتایج این آزمایش نشان میدهد که با توجه به تاثیر مثبت عملآوری گندم بر عملکرد رشد و قابلیت هضم مواد مغذی، با روشهای مناسب فرآوری میتوان به شکل موثرتر و با بازده بالاتر از گندم به عنوان منبع تامین کننده انرژی خوراک در تغذیه نشخوارکنندگان استفاده نمود. واژههای کلیدی: فرآوریهای مختلف گندم، قابلیت هضم، ترکیبات شیمیایی و بره پرواری

تعیین اهداف اصلاحی و ضرایب اقتصادی بز عدنی در سیستم پرورش مرتعی
چکیده سابقه و هدف: افزایش سوددهی هر واحد تولیدی، مهمترین راهکار برای افزایش قدرت رقابت آن در همه زمینهها و از جمله دامپروری میباشد. اولین قدم در طراحی برنامههای اصلاح نژاد دام، تصمیمگیری در مورد اهداف اصلاحی مناسب بوده و بزرگترین عامل در ناکارآمد بودن برنامههای اصلاح نژاد دام، عدم تبیین صحیح اهداف اصلاحی است. بنابراین تعیین اهداف اصلاحی و ضرایب اقتصادی صفات مهم در بز عدنی که یکی از دامهای مهم نوار ساحلی خلیج فارس است، کمک زیادی برای اصلاح نژاد این دام خواهد کرد. مواد و روشها: در این مطالعه از یک مدل ثابت قطعی که فرض میکند تنوعی در میان حیوانات برای صفات مهم مورد استفاده در محاسبه ضرایب اقتصادی وجود ندارد، استفاده گردید. به منظور برآورد پارامترهای مورد نیاز (جمعیتی، تولیدی، تولیدمثلی، مدیریتی و اقتصادی)، به مدت یک سال کامل تولید (از ابتدای شهریور 95 تا ابتدای شهریور 96)، تعداد 7 گله بز عدنی با 920 رأس بز مولد تحت رکوردگیری مستقیم قرار گرفتند. با تشکیل معادلات سود و تغییر صفات به اندازه یک درصد یا یک واحد، در حالی که میانگین سایر صفات در حد میانگین جمعیت ثابت بودند، ضرایب اقتصادی صفات محاسبه گردید. وزن اقتصادی صفات به ازاء یک انحراف معیار ژنتیکی افزایش در صفت مربوطه در حالی که سایر صفات در حد میانگین ثابت نگه داشته شدند، محاسبه شد. برای مقایسه صحیح ضرایب اقتصادی صفتها، اهمیت نسبی آنها تعیین شد. یافتهها: نتایج این تحقیق نشان داد که بیشترین درآمد، سهم فروش بزغاله و شیر و بیشترین هزینه سهم مدیریت و کارگری بود. صفاتی که جزء اهداف اصلاحی بز عدنی به دست آمدند شامل میزان بزغالهزایی، تعداد زایش در سال، تعداد بزغاله متولد شده در هر زایش، وزن فروش بزغاله در 12 ماهگی، میزان آبستنی، زندهمانی بزغاله تا شیرگیری، زندهمانی بز مولد، زندهمانی از شیرگیری تا فروش، شیر تولیدی، وزن بلوغ بزهای مولد و میزان دوقلوزایی با ضرایب اقتصادی به ترتیب 7/22، 5/17، 8/12، 1/12، 1/10، 8/9، 9، 6/4، 9/1، 7/0-، 3/0 بودند. نتیجهگیری: در بز عدنی صفات تولیدمثلی، ماندگاری و تولیدی به ترتیب دارای بیشترین اهمیت هستند. با توجه به شرایط حاکم بر سیستم پرورش این دام، صفات تعداد زایش در سال، میزان بزغالهزایی، تعداد بزغاله متولد شده در هر زایش، وزن فروش بزغاله در 12 ماهگی، میزان آبستنی، زندهمانی بزغاله تا شیرگیری، زندهمانی بز مولد، زندهمانی از شیرگیری تا فروش و شیر تولیدی به ترتیب از مهمترین صفات هستند و باید در برنامههای اصلاح نژاد بز عدنی مورد توجه بیشتری قرار گیرند.

تاثیر جایگاه فری استال بر میزان شیوع لنگش و شمار سلول بدنی شیر در گاوداری های صنعتی استان مازندران
سابقه وهدف: طی چند دههی اخیر با بکارگیری فناوریهای اصلاحنژادی، تغذیهای و مدیریتی، سطح تولید در گاوهای شیری بهطور قابلتوجهی افزایش یافته است. در این مدت مطالعات متعددی گزارش کردهاند که سطح تولید حیوان به شکل معنیداری تحت تاثیر رفاه و محل نگهداری حیوان میباشد. از جمله عواملی که تولید و اقتصاد گاوداریها را تحت تاثیر قرار میدهد، وجود یا عدم وجود ناهنجاریهای سیستم پستانی و حرکتی میباشد. اختلال در سطح آسایش و رفاه حیوان شانس ابتلا به این ناهنجاریها را افزایش می دهد. هدف از انجام پژوهش حاضر بررسی ارتباط بین میزان وقوع لنگش و شمار سلولهای بدنی موجود در شیر با سیستم پرورشی فریاستال یا غیر فریاستال در گاوداریهای صنعتی استان مازندران بوده است. مواد و روشها: در این پژوهش از رکوردهای زایش چهار گاوداری صنعتی واقع در استان مازندران با سیستم نگهداری فریاستال و غیر فریاستال طی دو سال 1393 و 1394 بهمنظور بررسی اثر جایگاه پرورشی بر نرخ ناهنجاری لنگش و شمار سلول بدنی شیر استفاده گردید. جهت بررسی ناهنجاری لنگش به دلیل ماهیت دودویی صفت از روش چند متغیره رگرسیون لجستیک و برای بررسی شمار سلول بدنی شیر از رگرسیون مختلط استفاده گردید. عواملی نظیر اثر گله، سال و فصل زایش، دوره شیرواری و جایگاه نگهداری بعنوان اثرات ثابت و سن در نخستین زایش در هر دو صفت و روزهای شیردهی در بررسی شمار سلولهای بدنی به عنوان متغیرهای کمکی در مدل در نظر گرفته شدند. یافتهها: تفاوت میانگین امتیاز سلول بدنی در جایگاههای فریاستال و غیرفریاستال معنیدار بود (05/0>P). بیشترین تفاوت امتیاز سلول بدنی بین دو جایگاه فریاستال و غیر فریاستال 23/0 ± 20/2 تخمین زده شد. همچنین شکم زایش اثر معنیدار بر امتیاز سلول بدنی داشته و با افزایش سن میزان آن بهطور متوسط از 99/1 به 2/3 رسید. اثر تمامی عوامل موجود در مدل بر میزان وقوع لنگش معنیدار بود (05/0>P). نتایج نشان داد در صورت استفاده از جایگاه فریاستال در مقایسه با جایگاه غیر فریاستال احتمال ابتلا به لنگش بهطور قابل ملاحظهای کاهش مییابد (16/2 – 52/1) 82/1) و درصد وقوع آن از 50/20 در جایگاههای غیر فریاستال به 58/13 در جایگاههای فریاستال کاهش یافت. نتیجهگیری: بر اساس نتایج به دست آمده میتوان گفت که استفاده از سیستم فریاستال در پرورش گاو شیری به میزان قابل توجهی نرخ وقوع ناهنجاری پستانی و حرکتی را کاهش میدهد. از این رو استفاده از این سیستم با رعایت اندازه و نسبت جایگاه به تعداد دام برای افزایش رفاه و سلامت دام میتواند به اقتصاد و سودآوری گلهها کمک قابل ملاحظهای نماید.

شناسایی چند شکلی در ناحیه تنظیمی بالادست ژن های کالپاین و کالپاستاتین و ارتباط آن با برخی صفات اقتصادی در گوسفندان لری بختیاری
سابقه و هدف: هدف از پژوهش حاضر بررسی تنوع آللی در ناحیه تنظیمی بالا دست ژنهای کالپاین و کالپاستاتین، تجزیه و تحلیل بیوانفورماتیک ژنهای مورد نظر و ارتباط آماری واریانتهای آللی با صفات وزن بدن و دنبه در گوسفند لری بختیاری بوده است. مواد و روش ها: تعداد 150 نمونه خون به طور تصادفی تهیه و DNA با روش نمکی بهینه یافته استخراج شد. سپس جفت آغازگرهای اختصاصی با نرم افزارOligo7 به ترتیب برای تکثیر قطعاتی با اندازههای 245 و 225 جفت بازی از ناحیه بالا دست ژنهای کالپاستاتین و کالپاین طراحی شدند. پس از انجام PCR فرآورده های تکثیری حاصل از جایگاه های نشانگری کالپاستاتین و کالپاین به ترتیب در معرض هضم با آنزیمهای اندونوکلئاز PstI و HaeIII قرار گرفتند. به دلیل یک شکل بودن نتایج حاصل RFLP، برای ارزیابی دقیق تر توالیهای تکثیر شده از تکنیک SSCP استفاده شد. بعد از تعیین ژنوتیپ، از هر الگوی باندی ژن کالپاین یک نمونه مورد توالی یابی قرار گرفت. برای شناسایی موتیفهای درگیر در فرآیند تنظیم ژنی و همچنین جهش های موجود در الگوهای باندی مختلف، توالیهای به دست آمده با استفاده از نرم افزارهای DNASIS MAX و BioEdit مورد بررسی قرار گرفتند. ارتباط الگوهای باندی مشاهده شده در جایگاه کالپاین با صفات وزن بدن در تولد، 3، 6 و 12 ماهگی و صفات دنبه شامل وزن، محیط بالا و پایین و ارتفاع دنبه در گوسفند لری بختیاری با استفاده از رویه Glimmix نرمافزار SAS (نسخه 1/9) مورد بررسی قرار گرفت. یافته ها: سه الگوی باندی مختلف A، B و C در جایگاه کالپاین در نمونه های لری بختیاری به ترتیب با فراوانی های 66، 9 و 25 در صد مشاهده شدند. ناحیه بالادست جایگاه ژنی کالپاستاتین در پژوهش حاضر یک شکل بود. تجزیه و تحلیل آماری وجود ارتباط آماری معنی داری را بین الگوهای باندی در جایگاه نشانگر کالپاین و صفت وزن تولد در گوسفندان لری بختیاری نشان داد (P

اثر جایگزینی دانه ی سویای فرآوری شده با کنجاله ی سویا بر تولید و ترکیب شیر، قابلیت هضم ظاهری مواد مغذی، فراسنجه های خونی و فراسنجه های تخمیری شکمبه گاوهای شیری هلشتاین در اواسط دوره ی شیردهی چکیده سابقه و هدف: فرآوری اقلام خوراکی گاه موجب افزایش بازده مصرف خوراک و نتیجتا عملکرد دام ها می گردد. در گاوهای شیری پر تولید نیاز به پروتئین بیش از ظرفیت تولیدی میروارگانیسم های شکمبه است. از این رو استفاده از منابع پروتئین عبوری، ضروری می باشد. فرآوری حرارتی سویا موجب افزایش محتوی پروتئین عبوری خوراک می شود که نتیجه ی آن افزایش جذب پروتئین جیره در روده ی باریک (با حداقل تغییرات) و به طبع آن افزایش بازده استفاده از نیتروژن خواهد بود. مواد و روش ها: از نه راس گاو اصیل هلشتاین چند شکم زایش در قالب طرح گردان با سه تکرار استفاده شد. برای هر دوره در هر تیمار 3 راس گاو در نظر گرفته شد. آزمایش در 3 دوره 21 روزه شامل 14 روز عادت پذیری به جیره های غذایی و 7 روز دوره جمع آوری، نمونه گیری و رکورد برداری، انجام شد. اندازه گیری عملکرد تولید شیر به صورت روزانه انجام شد. روزهای 19 و 20 هر دوره، نمونه گیری از شیر جهت تعیین چربی، پروتئین، لاکتوز و کل مواد جامد شیر به عمل آمد. نیتروژن خون، شکمبه و شیر در هر دوره نمونه گیری اندازه گیری شد. قاببلیت هضم مواد مغذی، اسیدیته ی شکمبه و فراسنجه های خونی گلوکز، نیتروژن اوره ای، پروتئین کل، تری گلیسیرید و کلسترول خون مورد مطالعه قرار گرفت. یافته ها: جایگزینی دانه ی سویای فرآوری شده با کنجاله ی سویا اثر معنی داری بر میزان مصرف ماده خشک و تغییرات وزن بدن نداشت. میزان کل تولید شیر، تولید شیر تصحیح شده بر اساس 3/5 درصد چربی، کل مواد جامد، درصد چربی، درصد پروتئین، درصد لاکتوز و نیتروژن شیر بین تیمارها تفاوت معنی داری نداشت. اسیدیته و نیتروژن آمونیاکی شکمبه تحت تاثیر تیمارها قرار نگرفت. قابلیت هضم ظاهری ماده ی خشک، ماده ی آلی، پروتئین خام و الیاف نامحلول در شوینده ی خنثی تحت تاثیر تیمارها قرار نگرفت. غلظت فراسنجه های خونی گلوکز، نیتروژن اوره ای ، پروتئین کل ، تری گلیسیرید و کلسترول بین گروه های مختلف آزمایشی یکسان بود. نتیجه: نتایج مطالعه ی حاضر نشان داد که جایگزینی کنجاله ی سویا با دانه ی سویای برشته شده و یا اکسترود شده منجر به بهبود عملکرد گاوهای هلشتاین اواسط شیردهی نمی شود. پس جایگزینی دانه ی سویای فرآوری شده با کنجاله ی سویا تنها در صورتی منطقی خواهد بود که قیمت سویای فرآوری شده از کنجاله ی سویا ارزان تر باشد و یا دانه ی سویا نسبت به کنجاله سویا، بیشتر در دسترس باشد. کلمات کلیدی: دانه سویا برشته شده، دانه ی سویای اکسترود شده، کنجاله سویا، گاو شیری، تولید شیر

شناسایی ریزRNAها، ژنهای هدف و مسیرهای سیگنالدهی مرتبط با تولید شیر با استفاده از miRNA-seq
سابقه و هدف: مولکولهای ریزRNA توالیهای کوتاهی (با میانگین طول 22 نوکلئوتید) هستند که با اثر بر تنظیم بیان ژنها فرآیندهای بیولوژیکی بسیاری را تحت تأثیر قرار میدهند. تولید شیر فرآیند فیزیولوژیکی میباشد که تحت تأثیر تعداد بسیار زیادی از ژنها، ریزRNAها، مسیرهای ژنی و مسیرهای سیگنالدهی قرار میگیرد. در این مطالعه با بررسی مولکولهای محافظت شده ریزRNA در بین گونههای موش، گاو و بز به شناسایی ریزRNAها، ژنهای هدف آنها و مسیرهای ژنی مرتبط با تولید شیر پرداخته شد. مواد و روشها: در این مطالعه جهت بررسی مکانسیم مولکولی اثر ریزRNAها بر تولید شیر، ابتدا دادههای مورد نظر با شماره دسترسی GSM1295115 برای گونه موش، GSM1295118 برای گونه گاو و GSM969927 برای گونه بز از پایگاه داده GEO دانلود شدند. شناسایی ریزRNAها با استفاده از نرمافزار mirdeep2 انجام شد. در این مطالعه از پایگاه داده mirwalk برای شناسایی ژنهای هدف ریزRNAهایی که دربافت پستان هر سه گونه بیان میشوند؛ استفاده گردید. پایگاه اطلاعاتی mirwalk قادر به تخمین ژنهای هدف بر اساس الگوریتمهای ده پایگاه اطلاعاتی دیگر میباشد. ترسیم ارتباطات ژنی شبکه برهمکنش ریزRNAها و ژنهای هدفشان توسط نرمافزار cytoscape انجام شد. جهت بررسی مسیرهای ژنی و سیگنالدهی مرتبط با ژنهای هدف از پایگاه اطلاعاتی DAVID استفاده شد. یافتهها: بر اساس نتایج این مطالعه ژنهای miR-93-5p، miR-27b-3p، miR-27a-3p و miR-200c-3p از مهمترین ریزRNAها و ژنهای Pten، Rlim، Pdik1l و Setd5 از مهمترین ژنهای هدف در فرآیند تولید شیر و مسیرهای بیوسنتزی ترکیبات شیر بودند. از مهمترین نتایج این مطالعه معرفی Setd5 به عنوان یک ژن جدید مرتبط با فرآیند تولید شیر میباشد. آنالیز مسیرهای ژنی نشان داد که مسیر چسبندگی کانونی، مسیر سیگنالدهیMAPK ، مسیر سیگنالدهی mTOR، مسیر سیگنادهی PI3K-Akt و مسیر سیگنالدهی نروتروفین از مهمترین مسیرهای ژنی میباشند که توسط ژنهای هدف فعال شده و نقش مهمی در بیوسنتز تولید شیر و توسعه بافت پستانی دارند. این مسیرهای ژنی میتوانند با اثرگذاری بر تکثیر سلولهای آلوئول، افزایش انشعابات، توسعه بافت پستانی، متابولیسم اسیدهای آمینه، اثر بر سیستم اندوکرینی، مسیرهای سیگنادهی پرولاکتین و سنتز ترکیبات شیر مانند چربی، پروتئین و لاکتوز، فیزیولوژی و بیولوژی تولید شیر را تحت تأثیر قرار دهند. نتیجهگیری: با توجه به نقش حیاتی ریزRNAهای مهم در شبکه مورد بررسی و ژنهای هدف این مولکولها و همچنین مسیرهای ژنی مورد مطالعه، میتوان در برنامههای اصلاح نژادی به عنوان مهمترین تنظیمکنندههای مربوط به فرآیند تولید شیر از آنها بهره برد. از این اطلاعات میتوان جهت معرفی و کاربرد ژنهای کاندید و کاربرد آنها در روش انتخاب به کمک ژن و یا انتخاب ژنومی استفاده کرد. با توجه به اینکه تولید شیر با توسعه و تکامل بافت پستانی همراه میباشد، مولکولهای ریزRNA و ژنهای هدفی که در این مطالعه به آن پرداخته شده است، میتوانند کاندید مناسبی در کلیه فرآیندهای تکامل و تمایز نیز مطرح گردند.

بررسی موقعیتهای جسم زرد و جنین و ارتباط آنها با فراسنجههای خونی میشهای آبستن و غیرآبستن لری بختیاری
سابقه و هدف: متابولیتهای مختلف از جمله گلوکز، کلسترول و نیتروژن اوره سرم از شاخصهای بسیار مهم برای بیان وضعیت پروتئین و انرژی در گوسفند هستند. نشان داده شده است که غلظت متابولیتهای سرم در زمانهای مختلف فعالیت تولیدمثلی و همچنین در میشهای آبستن با یک یا دو جنین تغییر میکند. بنابراین هدف از انجام این مطالعه بررسی تاثیر وضعیتهای مختلف تولیدمثلی بر غلظت متابولیتهای مختلف سرم در میشهای لری بختیاری بود. مواد و روشها: این تحقیق طی فصول تولیدمثل و غیر تولیدمثل در میشهای آبستن و غیر آبستن انجام شد. در ابتدای آزمایش با مراجعه به کشتارگاه، میشهای نژاد لری بختیاری با سن 3 الی 4 سال انتخاب شدند. سپس نمونههای خون از همه میشها قبل از کشتار جمعآوری و برای اندازهگیری گلوکز، کلسترول و نیتروژن اوره سرم به آزمایشگاه منتقل شدند. بخش اول این پژوهش در فصل غیر تولیدمثلی انجام شد و 48 عدد دستگاه تولیدمثلی غیرآبستن جمعآوری و به سه گروه تیماری بهصورت زیر تقسیم شدند. گروه 1: دارای یک جسم زرد؛ گروه:2 دارای دو جسم زرد (یکطرفه) و گروه 3: دارای دو جسم زرد (دو طرفه). بخش دوم این پژوهش طی فصل تولیدمثلی انجام شد. 144 دستگاه تولیدمثلی جمعآوری و در 9 گروه تیماری به صورت زیر تقسیم شدند. گروه 1: دارای یک جسم زرد و غیرآبستن؛ گروه 2: دارای دو جسم زرد (یکطرفه) و غیرآبستن؛ گروه 3: دارای دو جسم زرد (دو طرفه) و غیرآبستن؛ گروه 4: دارای یک جسم زرد با یک جنین؛ گروه 5: دارای یک جسم زرد با دو جنین؛ گروه 6: دارای دو جسم زرد (یکطرفه) با یک جنین؛ گروه 7: دارای دو جسم زرد (دوطرفه) با یک جنین؛ گروه 8: دارای دو جسم زرد (یکطرفه) با دو جنین و گروه 9: دارای دو جسم زرد (دو طرفه) با دو جنین. یافتهها: این پژوهش نشان داد که از بین تیمارهای آزمایشی، بالاترین غلظت گلوکز سرم در میشهای دارای یک جسم زرد با یک جنین و پائینترین غلظت گلوکز سرم در میشهای دارای دو جسم زرد (دو طرفه) با دو جنین مشاهده شد (05/0>P). همچنین غلظت گلوکز سرم در میشهای دارای دو جسم زرد (یک طرفه و دوطرفه) که تنها یک جنین داشتند نسبت به میشهای دارای یک جسم زرد با دو جنین بهطور معنیداری بیشتر بود (05/0>P). نتایج نشان داد که غلظت کلسترول سرم میشها در فصل تولیدمثل نسبت به فصل غیرتولیدمثل در هر دو وضعیت جسم زرد منفرد و دوتایی بهطور معنیداری بیشتر بود (05/0>P). علاوه بر این، غلظت کلسترول سرم در میشهای دارای یک جسم زرد و دو جنین نسبت به میشهای دارای جسم زرد دوتایی (دو طرفه) با دو جنین اختلاف معنیداری نداشت. غلظت نیتروژن اوره سرم نیز در میشهای غیر آبستن طی فصل تولیدمثل بهطور معنیداری (05/0>P) بیشتر از فصل غیر تولیدمثل بود. بهعلاوه، غلظت این متابولیت بین میشهای با موقعیتهای مختلف جسم زرد (یکطرفه و دوطرفه) و همچنین میشهای دارای یک جسم زرد و دو جنین اختلاف معنیداری نداشت. نتیجهگیری: نتایج بهطور کلی نشان داد که غلظت گلوکز سرم طی فصل غیر تولیدمثلی نسبت به فصل تولیدمثلی بیشتر بود در حالیکه در میشهای آبستن نسبت به میشهای غیر آبستن همچنین در میشهای با دو جنین نسبت به یک جنین کمتر بود. غلظت کلسترول و نیتروژن اوره سرم نیز در میشهای آبستن نسبت به میشهای غیر آبستن و در میشهای دارای دو جنین نسبت به یک جنین بیشتر بود.

اثر اسید لینولئیک مزدوج خوراکی و مکمل تزریقی سلنیم و ویتامین E برعملکرد تولیدی و فراسنجه های خونی گاوهای هلشتاین
چکیده سابقه و هدف: اسید لینولئیک مزدوج به گروهی از ایزومرهای هندسی و موقعیتی اسید لینولئیک 18 کربنه (2: 18C) اشاره دارد که با یک پیوند ساده از هم جدا شده اند. دو ایزومر شناخته شده این اسید، ایزومر سیس 9 ترانس 11 و ترانس 10 سیس 12 می باشند. مطالعات نشان داده است که ایزومر ترانس 10 سیس 12 چربی شیر را در گاوهای شیری کاهش می دهد. مکمل اسید لینولئیک مزدوج با کاهش انرژی مورد نیاز برای سنتز چربی شیر، بالانس انرژی را در گاوهای شیری بهبود می بخشد. از طرفی با بهبود روشهای محافظت کردن چربی ها از بیوهیدروژناسیون در شکمبه، استفاده از آنتی اکسیدانها هنگامی که چربی های غیر اشباع محافظت شده خورانیده مورد نیاز می باشد. سلنیم و ویتامین E مواد مغذی ضروری هستند که در نقش بیولوژیکی مهمی به عنوان آنتی اکسیدان سهیم هستند. هدف از آزمایش حاضر مطالعه تأثیر مکمل اسید لینولئیک مزدوج محافظت شده و مکمل تزریقی سلنیم و ویتامین E بر عملکرد تولیدی و فراسنجه های خونی در گاوهای شیرده هلشتاین در دوره انتقال و اوایل شیردهی بود. مواد و روشها: تعداد۴۰ راس گاو شیری بر اساس تولید شیر دوره قبلی و نوبت زایش انتخاب و از حدود 21 روز پیش از زایش تا 60 روز پس از آن، به طور تصادفی در بین 4 تیمار آزمایشی قرار گرفتند. تیمارهای آزمایشی شامل 1- جیره پایه (کنترل)2- جیره پایه بعلاوه مکمل تزریقی سلنیم و ویتامین E3- جیره پایه بعلاوه 100 گرم مکمل اسید لینولئیک مزدوج در روز 4- جیره پایه بعلاوه 100 گرم مکمل اسید لینولئیک مزدوج همراه مکمل تزریقی سلنیم و ویتامین E.بود. مکمل سلنیم و ویتامین E به میزان 40 سی سی در روزهای21-، 1، 14، 28 و 42 نسبت به روز زایش به صورت عضلانی به هر گاو تزریق شد. مقدار خوراک مصرفی و تولید شیر به صورت روزانه، ترکیبات شیر به صورت هفتگی، وزن و امتیاز بدنی با فواصل 21 روز تعیین شدند. نمونه های خون نیز از همه گاوها درروزهای تزریق گرفته شد. یافته ها: افزودن مکمل اسید لینولئیک مزدوج تأثیر معنی داری بر افزایش تولید شیر و بهبود بازده تولیدشیر و توازن انرژی داشت (01/0>(P). استفاده از مکمل اسید لینولئیک مزدوج درصد و مقدار چربی شیررا به طور معنی داری کاهش داد(001/0>P). اما تأثیرمعنی داری بر درصد پروتئین ،موادجامد بدون چربی و تعداد سلولهای سماتیک شیر نداشت (05/0P>). اگرچه میزان و مقدار پرونئین ،لاکتوز و مواد جامد بدون چربی شیررا افزایش داد (001/0>P). افزودن مکمل اسید لینولئیک مزدوج باعث بهبود غیر معنی داربرخی از صفات تولید مثلی مانند تعدادتلقیح به ازای آبستنی و روزهای بازغیرآبستنی شد. (05/0P>).. افزودن مکمل اسید لینولئیک مزدوج تأثیر معنی داری برغلظت فراسنجه های خون نداشت (0.05P>). اثرات متقابل اسید لینولئیک مزدوج و مکمل و سلنیم و ویتامین E بر غلظت گلوکز پلاسما معنی دار بود (0.05P<). گاوهای تغذیه شده با جیره شاهد کمترین غلظت گلوکز پلاسما را داشتنددر مقابل گاوهای تغذیه شده با جیره حاوی مکمل اسید لینولئیک مزدوج بیشترین غلظت گلوکز پلاسما را داشتند. نتیجه گیری: نتایج این پژوهش نشان داد استفاده از اسید لینولئیک مزدوج سبب کاهش چربی شیر و افت کمتر نمره وضعیت بدنی و افزایش تولید شیر و بهبود بالانس انرژی شده اما تأثیر معنی داری بر غلظت فراسنجه های خونی ندارد. استفاده از مکمل تزریقی سلنیم و ویتامین E سبب کاهش تولید چربی شیر و افزایش مواد جامد بدون چربی شیر می شود .استفاده از مکمل اسید لینو لئیک مزدوج و مکمل تزریقی سلنیم و ویتامین E سبب بهبود غیر معنی دار عملکرد تولید مثلی شده است.

بررسی تغییرات ترکیب شیمیایی، روند تجزیه پذیری شکمبهای و هضم رودهای نشاسته و پروتئین خام واریتههای مختلف دانه جو پرتوتابی شده با ریزموج
سابقه و هدف: پرتوتابی ریزموج نوعی عملآوری حرارتی برای کاهش تجزیهپذیری شکمبهای پروتئین خام، نشاسته و افزایش پروتئین عبوری به روده کوچک در جهت بهبود تخمیر شکمبهای غلات میباشد. این پژوهش در راستای بررسی روند تجزیهپذیری نشاسته و پروتئین خام ارقام مختلف دانه جو پرتوتابی شده با ریزموج و نحوه توزیع پروتئین در سیستم کربوهیدرات و پروتئین خالص کورنل و سیستم پروتئین قابل متابولیسم انجام شد. مواد و روشها: تعیین ترکیبات شیمیایی با آسیاب آزمایشگاهی با الک 1 میلیمتری آسیاب و غلظت ماده خشک، ماده آلی پروتئین خام (دستگاههای کجلدال )، چربی خام با استفاده از روشهای استاندارد AOAC(2000)، نشاسته به روش آنتروم (44) و الیاف نامحلول در شوینده خنثی (سیستم آنکوم ) و الیاف محلول در شوینده اسیدی با روش ون سوست و همکاران (1991) اندازهگیری (سه تکرار) شد. ارقام جو ماکویی، بهمن آبی و سهند بهمدت 2، 4 و 6 دقیقه در معرض تابش پرتو ریزموج (مایکرویو) قرار گرفتند. کنتیک و فراسنجههای تجزیهپذیری شکمبهای ماده خشک، نشاسته و پروتئین نمونههای خوراکی بهروش کیسههای نایلونی با استفاده از سه رأس گوساله نر نژاد هلشتاین مجهز به فیستولای قابل انعطاف شکمبهای در ساعات 0، 2، 4، 6، 8، 12، 24 و 48 ساعت و در قالب طرح بلوکهای کامل تصادفی تعیین شد. توزیع پروتئین بر اساس سیستم پروتئین قابل متابولیسم بر اساس معادلات مربوطه تعیین و میزان حلالیت پروتئین در بافرهای مختلف بهمنظور فهم نحوه توزیع بخشهای مختلف پروتئین در سیستم کربوهیدرات و پروتئین خالص کورنل (CNCPS) تعیین شد. علاوه بر این اثر پرتوتابی بر نحوه توزیع کربوهیدراتهای دانه جو در این سیستم مورد مطالعه قرار گرفت. یافتهها: تحت تأثیر پرتوتابی ریزموج میزان ماده خشک، پروتئین خام، مادهآلی، الیاف محلول در شوینده اسیدی، الیاف نامحلول در شوینده خنثی ارقام مختلف دانه جو کاهش یافت بهطوری که در مدت زمان 6 دقیقه کمترین میزان ماده خشک، پروتئین خام، مادهآلی، الیاف محلول در شوینده اسیدی ،الیاف نامحلول در شوینده خنثی و چربی خام مربوط به رقم سهند بود. نتایج این آزمایش نشاندهنده تأثیر کاهنده و معنیدار پرتوتابی در زمانهای مختلف بر میزان نشاسته و پروتئین قابل تجزیه مؤثر در شکمبه و وجود اختلاف معنیداری بین ارقام مختلف مورد بررسی از این نظر بود. در بین ارقام مختلف مورد مطالعه، فراسنجههای مختلف تجزیهپذیری رقم جوی سهند بیش از سایر ارقام تحت تأثیر عملآوری قرار گرفت. تفاوت بین ارقام مختلف در پاسخ به عملآوری را میتوان بازتابی از میزان پوسته، نشاسته و پروتئین خام و توزیع متفاوت نشاسته در بخشهای مختلف دانه دانست. افزایش زمان پرتوتابی سبب افزایش تأثیرگذاری آن بر فراسنجههای مختلف موردارزیابی در ارتباط با تجزیهپذیری پروتئین خام و نشاسته شد. عملآوری ارقام مختلف دانه جو با پرتوهای ریزموج سبب کاهش معنیدار میزان پروتئین محلول، کاهش نیتروژن غیرپروتئینی و افزایش در مقادیر پروتئین و نشاسته بالقوه قابل دسترس در سیستم کربوهیدرات و پروتئین خالص کورنل شد. بااینحال، افزایش زمان پرتوتابی سبب افزایش مقادیر پروتئین متصل به دیواره سلولی و افزایش معنیدار پروتئین غیرقابل دسترس شد ولی تأثیری بر مقادیر نشاسته قابل دسترس نداشت. برخلاف نتایج آزمون سیستم کربوهیدرات و پروتئین خالص کورنل، پرتوتابی تأٍثیر منفی بر گوارش پذیری پس شکمبهای پروتئین خام ارزیابی شده بر اساس روش سه مرحلهای آنزیمی نداشت ولی میزان گوارشپذیری نشاسته را تحت تأثیر قرار داد. نتیجهگیری: نتایج این مطالعه نشاندهنده تفاوت در پاسخ ارقام مختلف جو به پرتوتابی با امواج ریزموج و لزوم انجام آزمون درونتنی بود. بهنظر میرسد بر اساس نتایج ارائه شده، بهترین پاسخ در تأثیرگذاری بر فراسنجههای ارزش غذایی در راستای بهبود تخمیر شکمبهای، تجزیهپذیری پروتئین، نشاسته و افزایش مقادیر نشاسته و پروتئین گوارشپذیر ورودی به روده باریک را میتوان مربوط پرتوتابی بهمدت 4 دقیقه در ارقام مختلف دانست.

مطالعه تابع توزیع زنده مانی در بزغاله های کرکی راینی از تولد تا یکسالگی
سابقه و هدف: علیرغم تغییرات زیاد در کشاورزی به دلیل پیشرفتهای صنعتی، جهانی شدن و پیشرفتهای تکنولوژیکی، جمعیت بز در جهان افزایش یافته است. پرورش بز یکی از عناصر کلیدی دخیل در اقتصاد کشاورزانی دارد که در نواحی خشک و نیمه خشک دربرگیرنده بیشتر مساحت ایران زندگی میکنند. حدود 30 میلیون راس بز کرکی در سراسر جهان وجود دارد که 5/4 تا 5 میلیون راس از آنها در ایران پرورش داده میشوند. بز کرکی راینی مهمترین نژاد کرکی کشور بوده و در جنوب شرقی ایران گسترده است و برای تولید گوشت و کرک نگهداری می شود. تقریبا سه میلیون رأس از این نژاد در استان کرمان وجود دارد. در سالهای اخیر برنامه ملی اصلاح نژاد به منظور مطالعه و حفاظت از تنوع ژنتیکی این نژاد اجرا شده، ولی منجر به پیشرفت زیادی در سوددهی اقتصادی آن نشده است که یکی از دلایل آن میتواند فقدان یک شاخص انتخاب اقتصادی برای صفات پایه در برنامه اصلاح نژاد باشد. زنده مانی یکی از صفات مهم اقتصادی و از عوامل تاثیرگذار بر درآمدزایی گله ها است زیرا هر گونه مرگ و میر یا حذف زودهنگام دام درآمد دامدار را کاهش می دهد. بنابراین، ورود این صفت در برنامه های اصلاحی می تواند منجر به بهبود آن و در نتیجه بازدهی اقتصادی بیشتر شود. با توجه به اهمیت زنده مانی و اینکه تا کنون زنده مانی بزهای کرکی راینی مطالعه نشده است، هدف از این مطالعه بررسی تابع توزیع زنده مانی بزغاله های کرکی راینی از زمان تولد تا یکسالگی بود. مواد و روش ها: در اﻳﻦ ﭘﮋوﻫﺶ از رﻛﻮردﻫﺎی زﻧـﺪهﻣـﺎﻧﻲ ﺗﻌـﺪاد 3055 راس بزغاله (حاصل از 201 راس بز نر و 1309 راس بز ماده)، ﻣﺮﺑﻮط ﺑﻪ ﮔﻠﻪ اﻳﺴﺘﮕﺎه ﭘﺮورش و اﺻﻼح نژاد بز کرکی راینی واﻗﻊ در شهرستان بافت که ﻃﻲ ﺳﺎلﻫﺎی 1386-1372 ﺟﻤﻊآوری ﺷﺪه بودند، استفاده شد. توزیع فراوانی علل حذف و تابع توزیع زنده مانی بزغالهها تا سن یکسالگی با استفاده از نرم افزار R برآورد شد. یافته ها: میزان حذف بزغالهها از تولد تا سن یکسالگی برابر با 09/22 درصد میباشد ﻛﻪ مقدار آن در سه ماه اول، دوم، سوم و چهارم به ترتیب 2/11، 08/5، 17/2 و64/3 درصد می باشد. مهمترین دلایل حذف به ترتیب اولویت مازاد پرواری و مرگ در اثر بیماری بوده است. میزان زنده مانی تجمعی بزغالهها از تولد تا سن یکسالگی 91/77 درصد بود. ضریب تابعیت میزان زنده مانی از سن نشان داد ﻛﻪ روزانه 048/0 درﺻﺪ از زنده مانی بزغالهها ﻛﺎﻫﺶ می یابد. نتیجه گیری: ﺑﺎ ﺗﻮﺟﻪ ﺑﻪ ﺑﺎﻻﺗﺮ ﺑﻮدن ﻣﻴﺰان ﺗﻠﻔﺎت در ﺳﻪ ﻣﺎهه اول زﻧﺪﮔﻲ بزغالهﻫﺎ ﭘﻴﺸﻨﻬﺎد ﻣﻲﮔﺮدد در ﻧﺤﻮة مدیریت و ﺳﻴﺴﺘﻢ ﭘﺮورش بزغالهﻫﺎ تجدید نظر گردیده و در کنار آن بهبود ظرفیت ژنتیکی و لحاظ نمودن صفت زنده مانی در برنامه های اصلاح نژادی مدنظر قرار گیرد.
