بررسی اثر امتیاز وضعیت بدنی میش بر صفات وزن بدن و تعداد بره در هر زایش در یک گله گوسفند شال
سابقه و هدف: نمره وضعیت بدنی در گوسفند بستگی به میزان بافتهای ماهیچه و چربی زوائد افقی و عمودی مهرههای انتهایی کمر داشته و میزان چاقی حیوان زنده را ارزیابی میکند. این امتیازدهی ضمن کمک به کنترل وضعیت میش، با تشخیص فوری تغییرات وضعیت بدن راهنمایی در جهت تغییر در مدیریت گله بوده و در نتیجه منجر به بهبود در صفات تولید و تولیدمثلی و افزایش بازدهی میشود. هدف از تحقیق حاضر بررسی اثر امتیاز وضعیت بدنی میش بر صفات وزن بدن بره در سنین مختلف و تعداد بره در هر زایش بود. مواد و روشها: در این مطالعه، امتیاز وضعیت بدنی 147 رأس میش نژاد شال، 10 الی 12روز قبل از جفتگیری در یک واحد گوسفنداری در شهرستان تاکستان بررسی شد. میشها در محدوده سنی 1 تا 6 سال و دامنه وزنی 45 تا 75 کیلوگرم قرار داشتند و از نظر امتیاز وضعیت بدن به پنچ گروه با فاصله 1 واحد (1، 2، 3، 4 و 5) تقسیم شدند. تعیین نمره وضعیت بدنی، با استفاده از انگشتان دست و از طریق لمس زواید افقی و عمودی چهار عدد از مهرههای کمری پشت و دنده آخر انجام گرفت. به منظور بررسی اثر وضعیت بدنی میش بر صفات مورد مطالعه (وزن تولد، وزن شیرگیری، وزن 5 ماهگی و تعداد بره در هر زایش) از نرمافزار Minitab 14 استفاده شد. تجزیه و تحلیل داده ها با استفاده از رویه GLM و Logistic به ترتیب برای صفات وزن بدن و تعداد بره در هر زایش انجام شد. یافتهها: اثر جنس و تعداد بره در هر زایش بر وزن بدن برهها معنی دار بود و برههای نر و یک قلو وزن بیشتری در زمان تولد، شیرگیری و 5 ماهگی نسبت به برههای ماده و دوقلو داشتند (05/0P<). وزن میش اثر معنی داری بر وزن بدن برهها در سنین مختلف نداشت. اثر سن میش بر وزن تولد بره ها معنیدار بود (05/0P<)، هرچند این اثر بر وزن شیرگیری و 5 ماهگی معنی دار نبود. اثر وزن تولد برهها بر صفات وزن شیرگیری و وزن 5 ماهگی از نظر آماری معنیدار بود و برههای سنگینتر در زمان تولد، وزن شیرگیری و 5 ماهگی بیشتری داشتند (001/0P<). اثر امتیاز وضعیت بدنی میش بر وزن برهها در سنین مختلف و تعداد نتاج در هر زایش، معنیدار بود (05/0P<). برههای حاصل از میشهای با امتیاز بدنی 3 و بیشتر، وزن بدن بیشتری را در زمان تولد، شیرگیری و 5 ماهگی نشان دادند. همچنین بیشترین میزان دوقلوزایی در میشهایی با امتیاز بدنی 3 مشاهده شد. نتیجهگیری: با توجه به اثر امتیاز وضعیت بدنی میش بر میزان دوقلوزایی و وزن بدن برهها، برای دستیابی به عملکرد تولیدی بیشتر در نژاد شال پیشنهاد می شود حداقل امتیاز وضعیت بدن 3 برای میشها در زمان جفتگیری مد نظر قرار گیرد. برای میشهایی با وضعیت بدنی پایین تر می توان از فلاشینگ یا تغذیه کمکی به منظور بهبود عملکرد تولید و تولیدمثل استفاده نمود.

تاثیر استفاده از تفاله دانه انار و ال-کارنیتین در جیره بر کمیت و کیفیت شیر و فراسنجههای شکمبهای بز شیرده
استفاده از فراوردههای فرعی کشاورزی و صنایع غذایی موجب کاهش هزینههای پرورش دام میگردد، تفاله دانه انار یک فراورده فرعی بوده که میتوان در تغذیه دام استفاده کرد. همچنین برای بهبود استفاده از روغن موجود در دانهی انار که بیشتر آن را اسیدهای چرب بلند زنجیر تشکیل میدهند میتوان با استفاده از ال-کارنیتین که بر اکسیداسیون اسیدهای چرب موثر میباشد، برای افزایش بازده تولید شیر بهره گرفت. جهت بررسی تاثیر تفاله دانه انار حاوی روغن غیراشباع به همراه ال– کارنیتین، بر کمیت و کیفیت شیر بزهای شیرده نژاد مهابادی و بررسی فعالیت شکمبهای بزها این آزمایش طراحی گردید. مواد و روش ها تعداد 32 راس بز شیرده نژاد مهابادی با روزهای شیردهی 6 ± 45، تولید شیر روزانه 20/0 ± 05/1 کیلوگرم، میانگین سن 3 سال، وزن 1/4 ± 8/39 کیلوگرم که دارای یک بزغاله شیرخوار بودند. بزها بر اساس دادههای جمعآوری شده در دوره پیش آزمایش به طور تصادفی به 4 گروه آزمایشی 8 رأسی تقسیم و سپس با آرایش فاکتوریل2×2 با دو سطح صفر و 10 درصد تفاله دانه انار و صفر و 300 قسمت در میلیون ال-کارنیتین در کیلوگرم ماده خشک به مدت 6 هفته در آزمایش بودند. جیرههای آزمایشی شامل: 1) جیره بدون تفاله دانه انار و ال-کارنیتین، 2) جیره با 10 درصد تفاله دانه انار بدون ال-کارنیتین، 3) جیره بدون تفاله دانه انار با 300 قسمت در میلیون ال-کارنیتین و 4) جیره با 10 درصد تفاله دانه انار با 300 قسمت در میلیون ال-کارنیتین در کیلوگرم ماده خشک، بودند. با توجه به تغذیه بزها به صورت انفرادی، مقدار خوراک مصرفی هر بز به طور روزانه ثبت شد. شیر تولیدی روزانه هر بز با استفاده از دستگاه شیردوش در دو وعده اندازهگیری شد. نمونه گیری از مایع شکمبه، در آخرین روز هفته پایانی آزمایش و سه ساعت بعد از خوراکدهی نوبت صبح و با استفاده از لوله مری متصل به پمپ خلاء صورت گرفت. تجزیه و تحلیل دادهها با استفاده از نرم افزار SAS و رویه MIXED انجام شد. یافته ها نتایج حاصل از این آزمایش نشان داد که افزودن تفاله دانه انار و ال-کارنیتین به جیره تأثیری بر وزن نهایی، کل افزایش وزن، افزایش وزن روزانه و ماده خشک مصرفی نداشت و تغییر ضریب تبدیل غیر معنی دار بود. تولید شیر (گرم در روز) تحت تأثیر تیمارهای آزمایشی قرار نگرفت، اما تولید شیر تصحیح شده بر مبنای 4 درصد چربی، درصد چربی شیر، تولید چربی و نسبت چربی به پروتئین شیر با افزودن تفاله دانه انار به طور معنی دار افزایش یافت (05/0>P)، همچنین استفاده از مکمل ال-کارنیتین باعث افزایش معنیدار درصد پروتئین و لاکتوز شیر گردید (05/0>P). pH، تعداد پروتوزوآ، غلظت نیتروژن آمونیاکی و غلظت کل اسیدهای چرب فرار شکمبه تحت تأثیر تیمارهای آزمایشی قرار نگرفت (05/0P). نتیجه گیری نتایج این آزمایش نشان داد که استفاده از تفاله دانه انار و افزودن ال-کارنیتین به جیره باعث بهبود کیفیت شیر بز شدند. اگرچه استفاده همزمان ال-کارنیتین و تفاله دانه انار بر تولید شیر تاثیر معنی نداری نشان نداد، اما تاثیر متقابل ال-کارنیتین و تفاله دانه انار بر فعالیت شکمبه باعث تولید بیشتر استات گردید. با توجه به نتایج بدست آمده میتوان نتیجه گیری کرد که تفاله دانه انار را میتوان به عنوان محصول جانبی ارزان با بهبود تولید و ترکیب شیر بز و ال–کارنیتین را نیز به عنوان مکمل موثر بر مصرف چربی جیرهای و تامین بهتر انرژی دردسترس در تغذیه بزهای شیری به کار برد.

برآورد اجزای واریانس وزن بدن گوسفند مرینوس در تولد و شیرگیری با استفاده از نشانگرهای تک نوکلئوتیدی و دو رویکرد حداکثر درستنمایی محدود شده و بیزی
مقدمه: برآورد دقیق اجزای واریانس ژنتیکی و غیر ژنتیکی با اطلاعات شجرهای و ژنومی، از ملزومات پیشبینی صحیح ارزشهای اصلاحی میباشد. دسترسی به آرایههای چندشکلی تکنوکلئوتیدی (SNP) با تراکم بالا و افزایش تعداد حیوانات با اطلاعات ژنوتیپی، دقت و صحت برآوردهای مبتنی بر جمعیت را افزایش میدهد. انتخاب ژنومی بهطور بالقوهای قادر است بیشتر واریانس ژنتیکی را توسط نشانگرها توجیه نماید. هدف از مطالعه حاضر، برآورد مؤلفه واریانس ژنتیکی افزایشی برای صفات وزن تولد و وزن شیرگیری در گوسفند مرینوس با دو روش حداکثر درستنمایی محدود شده و بیزی بود. مواد و روشها: برای انجام این پژوهش از اطلاعات گوسفندان مرینوس استرالیایی که با تراشه نشانگری SNP50k شرکت ایلومینا، تعیین ژنوتیپ شده بودند، استفاده شد. پس از کنترل کیفیت دادههای فنوتیپی و نشانگری، 2189 فرد و 45875 نشانگر برای انجام تجزیه و تحلیل استفاده شدند. صفات مورد بررسی در این تحقیق، وزن تولد (1331 رکورد) و وزن شیرگیری (2136 رکورد) بودند. برای مطالعه رابطه بین فراوانی آللی و مقدار واریانس ژنتیکی افزایشی توجیه شده، SNPها در پنج گروه مختلف از فراوانی آللی کمیاب (MAF)، با تعداد تقریبا برابر در هر گروه، طبقه بندی شدند (18/0-0، 28/0-18/0، 36/0-28/0، 43/0-36/0 و 499/0-43/0). تجزیه و تحلیل با دو رویکرد حداکثر درستنمایی محدود شده (REML) و بیزی با استفاده از تکنیک نمونهگیری گیبس و مدل RKHS انجام گرفت. یافتهها: مقدار وراثتپذیری ژنومی برآورد شده با همه SNPها در رویکر REML برای وزن تولد و وزن شیرگیری به ترتیب برابر 07/0±58/0 و 05/0±46/0 بود. این مقدار وراثتپذیری در آنالیز بیزی و به روش RKHS برای صفات مذکور به ترتیب برابر 07/0±58/0 و 05/0±46/0 برآورد شد. برآوردهای به دست آمده از 5 گروه مختلف MAF، در آنالیزهای جداگانه و توأم، با هم متفاوت بود. برای هر دو صفت، در دو رویکرد REML و بیزی، مقادیر واریانس ژنتیکی افزایشی در تجزیههای جداگانه، برای همه گروهها، بیشتر از مقادیر به دست آمده در آنالیز توأم بود. در تجزیه و تحلیل مجزای گروههای مختلف MAF، در هر دو رویکرد ، مقدار وراثتپذیری ژنومی به دست آمده، برای گروههای مختلف، مشابه بود ولی در تجزیه توأم، بین دو رویکرد REML و بیزی مقدار واریانس ژنتیکی توجیه شده در زیرگروههای مختلف MAF، تفاوت زیادی وجود داشت. در رویکرد REML در تجزیه و تحلیل توأم، مقدار وراثتپذیری برای گروه 2 (28/0-18/0MAF=) در وزن تولد و برای گروه 5 (499/0-43/0MAF=) در وزن شیرگیری مقدار صفر به دست آمد. در رویکرد Bayes برای هیچکدام از گروهها، مقدار وراثتپذیری برابر صفر نبود. در مجموع واریانسهای ژنتیکی پنج گروه مختلف MAF، که در آنالیز جداگانه نسبت به واریانس محاسبه شده به وسیله همه SNPها به صورت همزمان، بسیار بزرگتر بود. اما مجموع این واریانسها در آنالیز توأم، مشابه مقدار به دست آمده از کل SNPها، برای هر دو صفت و در هر دو رویکرد بود. نتیجهگیری: در رویکرد بیزی یک توزیع پیشین مشترک برای واریانسها در نظر گرفته میشود، بنابراین به نظر میرسد نتایج حاصل از رویکرد بیزی دقیقتر و معتبرتر از رویکرد دیگر باشد. اگر چه تعداد SNPها در گروههای مختلف، مشابه بود، اما مقدار واریانس ژنتیکی توجیه شده توسط گروههای مختلف MAF متفاوت بود.

اثر سطح کربوهیدراتهای غیر فیبری و نوع غله بر عملکرد رشد، گوارش پذیری، خصوصیات لاشه و اندام-های درونی برههای پرواری نژاد لری
سابقه و هدف: غلات مختلفی در تغذیه گوسفند استفاده میشوند. این غلات سطوح و ترکیبات متفاوتی از کربوهیدرات های غیرفیبری را دارند که می تواند قابلیت هضم جیره و در نتیجه عملکرد بره را تحت تاثیر قرار دهند. لذا هدف از این پژوهش بررسی تأثیر سطح کربوهیدراتهای غیر فیبری جیره و نوع دانهی غله بر عملکرد، گوارش پذیری مواد مغذی و خصوصیات لاشه برههای پرواری بود. مواد و روشها: برای اجرای این پژوهش، بیست بره پرواری نر با میانگین سنی90 روز و میانگین وزن 1/1±25 کیلوگرم در قالب آزمایش فاکتوریل بر پایه طرح کاملاً تصادفی با چهار تیمار و پنج تکرار به مدت 80 روز آزمایش شدند. تیمارهای آزمایشی شامل 1-تیمار حاوی دانه جو آسیاب شده و 33 درصد کربوهیدرات غیر فیبری، 2- تیمار حاوی دانه جو آسیاب شده و 40 درصد کربوهیدرات غیر فیبری، 3- تیمار حاوی دانه ذرت آسیاب شده و 33 درصد کربوهیدرات غیر فیبری و 4- تیمار حاوی دانه ذرت آسیاب شده و 40 درصد کربوهیدرات غیر فیبری بودند. تیمارهای آزمایشی از لحاظ انرژی قابل متابولیسم، پروتئین خام، سطح علوفه به کنسانتره و میزان دیواره سلولی یکسان بودند. در این آزمایش نمونهگیری خوراک و مدفوع در طول دوره در دو نوبت انجام و در پایان دوره پروار، برای اندازهگیری خصوصیات لاشه از هر تیمار 2 رأس کشتار شده و پارامترهای مورد آزمون تعیین گردید. یافتهها: نتایج این پژوهش نشان داد که با افزایش سطح کربوهیدرات غیر فیبری، گوارش پذیری ماده آلی، فیبر نامحلول در شوینده خنثی و خاکستر خام کاهش یافت. استفاده از دانه ذرت باعث افزایش گوارش پذیری ماده آلی، فیبر نامحلول در شوینده اسیدی و چربی خام شد. همچنین تیمارها از لحاظ وزن لاشه، درصد ران و درصد لاشهی بدون دنبه و درصد چربی بطنی و دنبه به لاشهی گرم با هم اختلاف معنیداری داشتند. کمترین درصد وزن کلیه و پانکراس مربوط به تیمار ذرت با40 درصد کربوهیدرات غیر فیبری بود. استفاده از ذرت با 33 درصد کربوهیدرات غیر فیبری سبب کاهش درصد شکمبه نگاری خالی شد. نتیجه گیری: نتایج این آزمایش نشان داد که عملکرد برهها، وزن لاشه، درصد ران و درصد لاشهی بدون دنبه و همچنین درصد چربی بطنی و دنبه به لاشهی گرم تحت تأثیر نوع دانه غلات قرار گرفت. گوارش پذیری ماده خشک تحت تأثیر نوع دانه غله نگرفت، اما گوارش پذیری ماده آلی در برههایی که از جیره حاوی ذرت تغذیه کردند، بیشتر بوده و به نظر می رسد تیمار ذرت با کربوهیدرات 40 درصد در بین تیمارها عملکرد بهتری داشته و برای برنامههای پرورشی سفارش میشود.

اثر سطوح مختلف کنسانتره جیره بر عملکرد رشد، مصرف خوراک و ترکیب بافت لاشه برههای نر پرواری شال
سابقه و هدف: پرواربندی بره یکی از منابع مهم تولید گوشت قرمز در کشور است. این حرفه با روشهای مختلف تغذیهای مثل چرا در مراتع، تغذیه در سیستم بسته با استفاده از مواد خوراکی پر انرژی و یا تلفیقی از هر دو روش انجام میشود. در حال حاضر به دلیل محدودیت ظرفیت مراتع کشور، استفاده از سیستم بسته پرواربندی با تغذیه سطوح بالای مواد کنسانترهای رایجتر شده است. این آزمایش با هدف تعیین اثر نسبتهای مختلف علوفه به کنسانتره جیره بر عملکرد رشد، مصرف و بازده خوراک و ترکیب بافت لاشه برههای نر پرواری شال در مؤسسه تحقیقات علوم دامی کشور انجام شد. مواد و روشها: تعداد 80 رأس برهی نر 5-4 ماهه نژاد شال با میانگین وزن زنده9/3 ±5/33 کیلوگرم در قالب یک طرح آزمایشی کاملاً تصادفی شامل چهار تیمار، چهار تکرار برای هر تیمار و پنج مشاهده در هر تکرار مورد استفاده قرار گرفت. چهار جیره با نسبتهای 30:70، 50:50، 70:30 و 90:10 علوفه به کنسانتره به ترتیب با 23/2، 43/2، 58/2 و 71/2 مگاکالری در کیلوگرم انرژی قابل متابولیسم و 143، 152، 161 و 170 گرم در کیلوگرم ماده خشک، پروتئین خام به طور تصادفی به چهار گروه بره اختصاص داده شد. جیرهها به شکل حبهشده تهیه گردید و روزانه سه نوبت در ساعات 8، 14 و 20 تا حد اشتها در اختیار برهها قرار داده شد. برهها در انتهای آزمایش بعد از 16 ساعت پرهیز غذایی، توزین شده و از هر تکرار یک بره کشتار شد. پس از کشتار و پوست کنی، اندامهای بدن مثل پوست، کله و پاها از بدن جدا شده و توزین گردید. اندامهای داخلی لاشهها و اعضاء دستگاه گوارش جدا شده و توزین گردید. لاشه پس از توزین، به مدت 24 ساعت در سردخانه با دمای 4 درجهی سانتیگراد نگهداری شده و سپس به دو نیم لاشه تقسیم گردید. نیم لاشه راست به شش قطعه اصلی گردن، سردست، قلوهگاه، راسته، ران و دنبه تفکیک شد و به جز دنبه، سایر قطعات مورد تفکیک بافتی قرار گرفته و بافتهای چربی بین عضلانی، چربی داخل عضلانی، گوشت لخم و استخوان آنها جدا شده و توزین گردید. یافتهها: براساس نتایج به دست آمده با افزایش سطوح کنسانتره در جیره، ماده خشک مصرفی و ضریب تبدیل خوراک به صورت خطی کاهش یافت (01/0P<). افزایش وزن روزانه با افزایش سطوح کنسانتره تا تیمار سوم افزایش و پس از آن کاهش خطی نشان داد (01/0P<). وزن کشتار، لاشه گرم و سرد، وزن زنده بدن خالی، گوشت لخم، استخوان، چربی زیرجلدی، چربی بین عضلات، مجموع چربی لاشه، نسبت گوشت لخم به چربی، ضخامت چربی پشت، سطح مقطع عضله راسته، قطعات گردن، سردست، سرسینه، راسته و ران تحت تأثیر جیرههای آزمایشی قرار نگرفت اما افزایش سطوح کنسانتره سبب کاهش خطی محتویات دستگاه گوارش و افزایش خطی درصد لاشه برههای پرواری شد (01/0P<). نتیجهگیری: این مطالعه مشخص نمود افزودن کنسانتره به جیره برههای نر پرواری شال تا 700 گرم در کیلوگرم ماده خشک، سبب بهبود ضریب تبدیل خوراک، عملکرد رشد و درصد لاشه، بدون اثر منفی بر نسبت گوشت لخم به چربی لاشه گردید.

بررسی آزمایشگاهی تأثیر فوزالون و دیازینون بر فراسنجههای تخمیری و برخی جمعیتهای میکروبی شکمبه
سابقه و هدف: آفتکشهای ارگانوفسفره مانند فوزالون و دیازینون، شامل گروه متنوعی از مواد شیمیایی بوده که علیه طیف وسیعی از آفات کشاورزی بکار برده میشوند. حیوانات نشخوارکننده در محیطی پرورش داده میشوند که میتوانند در معرض این آفتکشها قرار بگیرند. مواجهه این حیوانات با این آفتکشها ممکن است عمدی (مانند استعمال پوستی برای کنترل حشرات و آلودگیهای پوستی) و یا تصادفی (مصرف خوراک آلوده به آفتکشها) باشد. اولین نگرانی مربوط به اثرات متغیر و بالقوه این مواد شیمیایی بر سلامتی دام و دومین نگرانی مخصوصاً برای تولیدکنندههای فرآوردههای دامی ایناست که این فرآوردهها به مواد شیمیایی آلوده نشده باشند. بنابراین هدف از این پژوهش بررسی سطوح مختلف فوزالون و دیازینون بر فراسنجههای تخمیری، جمعیت پروتوزوآ و یک گونه باکتری فیبرولیتیک (بوتیریویبریو فیبریسولونس: (Butyrivibrio fibrisolvens با روش Real Time PCR بود. مواد و روشها: در این پژوهش از دو نوع آفتکش ارگانوفسفره فوزالون و دیازینون در سه سطح (0، 100 و 500 پی پی ام) و بنتونیت سدیم در دو سطح (0 و 2 درصد مادهخشک جیره) در یک محیط کشت آزمایشگاهی و در یک آزمایش فاکتوریل 2×3×2 در قالب طرح کاملاً تصادفی استفاده شد. در ادامه تولید گاز اندازهگیری شد و برخی از پارامترها بر اساس روش تولید گاز، تخمین زده شد. همچنین کل جمعیت پروتوزوآ و گونه باکتری بوتیریویبریو فیبریسولونس در محیط کشت با روش Real Time PCR تعیین شدند. یافتهها: افزودن فوزالون یا دیازینون به محیط کشت، منجر به کاهش معنیدار تولید گاز تجمعی در زمان 12، 24 و 48 ساعت انکوباسیون و نیز نرخ (cgas) و پتانسیل تولید گاز (bgas) شد. همچنین کلیة مؤلفههای تخمین زده شده از قبیل قابلیت هضم ماده آلی (OMD)، انرژی قابل متابولیسم (ME)، انرژی خالص برای شیردهی (NEl)، تولید پروتئین میکروبی (MPY) و اسیدهای چرب کوتاه زنجیر (SCFA) در اثر افزودن فوزالون و دیازینون به محیط کشت، بهطور معنیداری کاهش یافت. اضافه کردن بنتونیت سدیم در سطح دو درصد به محیط کشت نیز منجر به کاهش معنیدار کلیة موارد ذکر شده در بالا گردید. با اضافه کردن هر یک از آفتکشها به محیط کشت، جمعیت پروتوزوآ کاهش معنیدار نشان داد ولی جمعیت گونه باکتری بوتیریویبریو فیبریسولونس تحت تأثیر مصرف دو آفتکش قرار نگرفت. کاربرد بنتونیت سدیم در محیط کشت منجر به کاهش معنیدار جمعیت پروتوزوآ و گونه باکتری بوتیریویبریو فیبریسولونس شد. نتیجهگیری: مصرف هر دو آفتکش فوزالون و دیازینون اثرات منفی بر تولید گاز، مؤلفههای تخمینی و جمعیت کل پروتوزوآ داشت. اثرات سمی فوزالون نسبت به دیازینون بر محیط کشت بیشتر بود. همچنین بنتونیت سدیم علاوه بر توکسین بایندر بودنش، نتوانست از اثرات منفی آفتکشهای دیازینون و فوزالون بر محیط کشت بکاهد و باعث ایجاد اثرات منفی بر تولید گاز، مؤلفههای تخمینی، گونه باکتری بوتیریویبریو فیبریسولونس و جمعیت کل پروتوزوآ شد.

اثر مکمل رومینوبافر، بیکربناتسدیم و مخلوط گیاهان داروئی بر اسیدوز ،اسیدهای چرب فرار، فراسنجه های خونی و عملکرد برههای پرواری کردی
سابقه و هدف: اسیدوز یکی از مشکلات اکثر دامداریها است که همه ساله خسارات مالی فراوانی به دامداریها تحمیل مینماید. با افزایش کنسانتره جیره و افزایش اسیدیته شکمبه، قابلیت هضم فیبر کاهش مییابد و تغییر در جمعیت میکروبی شکمبه مشاهده خواهد شد. جیرههایی که دارای سطوح بالایی از کنسانتره هستند ممکن است با اختلال در مکانیسم گوارش منجر به کاهش ظرفیت بافری شکمبه شوند، متعاقباً اسیدیته شکمبه را تغییر دهند که منجر به اسیدوز میشود. مطالعات نشان دادهاند استفاده از بافرها در تغذیه دام، شرایط محیطی شکمبه را بهبود بخشیده و بوسیله تعدیل اسیدیته شکمبه، از افت pH جلوگیری مینماید. مکمل رومینوبافر به منظور پیشگیری از اسیدوز و بهبود عملکرد دام از ترکیب بافرهای رایج و گیاهان دارویی تولید شده است. تحقیق حاضر جهت مقایسه اثر مکمل رومینوبافر، بیکربنات سدیم و مخلوط گیاهان داروئی بر پیشگیری از اسیدوز و بهبود عملکرد دامهای پرواری انجام شد. مواد و روشها: سی و شش رأس بره نر نژاد کردی پنج ماهه با وزن اولیه 2±25 کیلوگرم به مدت 100 روز به طور تصادفی در چهار گروه 9 راسی مورد بررسی قرار گرفتند و بصورت انفرادی نگهداری شدند. تیمارها شامل گروه 1: تیمار شاهد (فقط جیره پایه بدون افزودن مکمل دریافت نمودند)؛ گروه 2: رومینوبافر (دو درصد از جیره پایه)؛ گروه 3: بیکربنات سدیم (یک درصد از جیره پایه) و گروه4: مخلوط گیاهان داروئی (دو درصد از جیره پایه)، مورد مطالعه قرار گرفتند. در طول دوره آزمایش؛ عملکرد برهها، pH شکمبه، میزان ابتلا به اسیدوز، نمونهبرداری از فراسنجههای شکمبهای و فراسنجههای خونی انجام گرفت. برای تعیین دامهای مبتلا به اسیدوز، ابتدا با مشاهده علائم ظاهری همچون کم اشتهایی، اسهال و دانههای هضم نشده در مدفوع، بعنوان دام ابتلا به اسیدوز شناسایی میشد سپس pH مایع شکمبه دام مورد نظر در ساعت های دو، چهار، شش و هشت بعد از خوراک دهی مورد بررسی قرار میگرفت. یافتهها: اثر تیمارهای آزمایشی بر اضافه وزن روزانه، خوراک مصرفی و ضریب تبدیل معنیدار بود (01/0.(P< اثر تیمارهای آزمایشی در جیرههای پر کنسانتره بر عدم بروز اسیدوز معنیدار بود (01/0P< و 55/52χ2=). مکمل رومینوبافر و بیکربنات سدیم از کاهش pH شکمبه در طول دوره پرواربندی جلوگیری نمودند (05/0.(P< اثر تیمارهای آزمایشی بر فراسنجههای شکمبهای از قبیل کل اسیدهای چرب فرار، اسید استیک، اسید پروپیونیک، اسید بوتیریک، نسبت استیک به پروپیونیک، نیتروژن آمونیاکی شکمبه و آنزیم TAC شکمبه مثبت و معنیدار بود (05/0>P). اثر تیمارهای آزمایشی بر سطح گلوکز، پروتئین تام، نیتروژن اورهای خون، کلسترول، بتا هیدورکسی بوتیرات، آنزیم TAC سرم خون در طول دوره آزمایش معنیدار بود (05/0>P). نتیجهگیری: با توجه به نتایج این آزمایش مکمل رومینوبافر سبب بهبود عملکرد و کاهش هزینه تولید در پرواربندی بره شد، بنابراین استفاده از مکمل رومینوبافر تا سطح دو درصد جیرههای پرواری پیشنهاد میشود.

مقایسه ارزش غذایی و خصوصیات تخمیر شکمبهای سیلاژ تاج خروس سبز (آمارانتوس هیپوکوندریاسوس) با سیلاژ ذرت
سابقه و هدف: امروزه کشت گیاهان علوفهای مقاوم و سازگار به شرایط خشک و کم آب در ایران از اهمیت خاصی برخوردار است. بر این اساس، اخیراً گیاه تاج خروس بهعنوان یک گیاه زراعی از خارج از کشور وارد الگوی زراعی شده است. از آنجا که تحقیقات بسیار اندکی در مورد امکان استفاده از این گیاه بهعنوان خوراک دام انجام گرفته است، لذا هدف از مطالعه حاضر بررسی امکان سیلو کردن این گیاه و مقایسه آن با سیلاژ ذرت بود. مواد و روشها: علوفه تاج خروس سبز و ذرت پس از برداشت به قطعات 5-3 سانتیمتری خرد شدند. به منظور تهیه سیلاژ، هر دو علوفه بهطور جداگانه در داخل لولههای پلیاتیلن بهطول 75 و قطر 16 سانتیمتر (در 4 تکرار) سیلو شدند. پس از گذشت زمانهای صفر، 40 و 60 روز پس از سیلو کردن، درب سیلوها باز شد و از آنها نمونهبرداری صورت گرفت. ترکیب شیمیایی، pH، خصوصیات شیمیایی (کربوهیدراتهای محلول در آب، نیتروژن آمونیاکی و غلظت اسیدهای چرب فرار)، قابلیت هضم (بهروش برونتنی) و کینتیک تخمیر شکمبهای (بهروش آزمون تولید گاز) هر دو سیلاژ در زمانهای صفر، 40 و 60 روز پس از سیلو کردن تعیین شدند. یافتهها: نتایج نشان داد که درصد ماده خشک و پروتئین خام علوفه تاج خروس در تمام زمانها بهطور معنیداری بیشتر از علوفه ذرت بود (05/0P<). تفاوت درصد ترکیبات شیمیایی (ماده خشک، ماده آلی، خاکستر خام، پروتئین خام، دیواره سلولی و دیواره سلولی فاقد همی سلولز) در هر دو علوفه در زمان صفر با سایر زمانها (40 و 60 روز) نیز معنیدار بود (05/0P<). مقدار pH و نیتروژن آمونیاکی در علوفه تاج خروس در هر 3 زمان نمونهبرداری بیشتر از علوفه ذرت بود (05/0P<). اما درصد کربوهیدراتهای محلول در آب در علوفه تاج خروس کمتر از علوفه ذرت بود (05/0P<). همچنین، سیلو کردن (اثر زمان) باعث کاهش معنیدار pH و کربوهیدراتهای محلول در آب در هر دو علوفه در فاصله زمان بین صفر تا 40 روز شد (05/0P<). غلظت اسید استیک، اسید پروپیونیک و کل اسیدهای چرب فرار در هر دو زمان 40 و 60 روز پس از سیلو کردن، در علوفه تاج خروس بهطور معنیداری بیشتر از علوفه ذرت بود (05/0P<). نتایج مربوط به تعیین قابلیت هضم نشان داد که درصد قابلیت هضم ماده خشک (DMD)، درصد قابلیت هضم ماده آلی (OMD) و محتوای ماده آلی قابل هضم (DOMD) در روز صفر در هر دو نوع علوفه یکسان بودند. اما درصد DMD در علوفه تاج خروس در روزهای 40 و 60 پس از سیلو کردن، نسبت به علوفه ذرت افزایش معنیداری نشان داد (05/0P<). بر اساس نتایج آزمون تولید گاز، حداکثر ظرفیت تولید گاز (b) و سرعت تولید گاز (c) در هر سه زمان نمونهبرداری در علوفه تاج خروس بهطور معنیداری با علوفه ذرت تفاوت داشت (05/0P<). همچنین، سیلو کردن (اثر زمان)، باعث کاهش معنیدار پارامترهای فوق شد. نتیجهگیری: بهطور کلی، با توجه به سطح مناسب پروتئین خام، اجزای دیواره سلولی و نیز قابلیت هضم علوفه تاج خروس و خصوصیات سیلویی آن از قبیل pH، میزان کربوهیدراتهای محلول در آب، این علوفه میتواند بهعنوان یک سیلاژ با کیفیت در تغذیه دام مورد استفاده قرار گیرد.

بررسی تغذیه برگ برهان بر تخمیر، تولید گاز، هضمپذیری و جمعیت پروتوزوای شکمبه شتر تک کوهانه
سابقه و هدف: برهان گیاهی از تیرهی بقولات بوده؛ بومی آفریقا، آسیا، بخش هایی از امریکا و استرالیا است و در ایران در استانهای خوزستان، بوشهر، فارس و هرمزگان کاشته میشود. برگ، غلاف و گل برهان برای نشخوارکنندگان خوراکهایی با ارزش هستند. بنابراین این آزمایش به منظور بررسی میزان تولید گاز، قابلیت هضم آزمایشگاهی و تعداد پروتوزوآ در شتر تک کوهانه تغذیه شده با جیره های حاوی برگ برهان جهت امکان جایگزینی آن حتی تا 100 درصد جایگزین یونجه انجام گرفت. مواد و روشها: در این آزمایش برگ های برهان از ملاثانی اهواز جمع آوری گردید. چهار شتر تک کوهانه 5 ساله با وزن25±300 کیلوگرم از نژاد عربی انتخاب شدند و به مدت 1 ماه با جیرهای با نسبت 60 درصد باگاس و 40 درصد یونجه (جیرهی بدون برگ برهان) و جیرهی حاوی برگ برهان (100 درصد جایگزین یونجه) که هر جیره دو تکرار داشت برای یک دورهی یک ماهه تغذیه شدند. در پایان دورهی آزمایش از شترهای تغذیه شده با جیرههای آزمایشی مایع شکمبه گرفته شد و شمارش تعداد پروتوزوآ، تولید گاز و قابلیت هضم آزمایشگاهی ماده فیبری و پروتئینی انجام گردید. یافتهها:بر اساس نتایج، هیچگونه پروتوزوآیی در مایع شکمبه شترهای تغذیه شده با جیرههای آزمایشی مشاهده نشد. تفاوت معنی-داری در پتانسیل تولید گاز و نرخ تولید مربوط به شترهای تغذیه شده با جیره های آزمایشی مشاهده شد (05/0>(P. پتانسیل تولید گاز باگاس و کنجاله سویا در شترهای تغذیه شده با جیره حاوی برهان بیشتر از جیره بدون برهان بود. فراسنجههای تخمیری مربوط به باگاس از جمله مادهی آلی واقعاً هضم شده و تودهی میکروبی در شترهای تغذیه شده با جیره حاوی برهان بیش از جیره بدون برهان بود (05/0>P )، همچنین راندمان سنتز تودهی میکروبی جیره برهان کمتر از جیره بدون برهان بود (05/0>P ). مقدار عامل تفکیک مربوط به جیره بدون برهان ( 10/10میلی گرم بر میلی لیتر) بالاتر از جیره حاوی برگ برهان (81/8 میلی گرم بر میلی لیتر) به دست آمد. همچنین فراسنجههای تخمیری مربوط به کنجاله سویا شامل مادهی آلی واقعاً هضمشده، تودهی میکروبی و راندمان سنتز تودهی میکروبی در جیره بدون برهان بیشتر از جیره حاوی برهان بود (05/0>P). عامل تفکیک نیز در گروه برهان و بدون برهان اختلاف معنیداری با هم نداشتند (05/0 نتیجهگیری کلی: بر اساس نتایج، هیچ گونه پروتوزوآیی در مایع شکمبه شترهای تغذیه شده با جیره های بدون برهان و حاوی برگ برهان مشاهده نشد و با جایگزین کردن 100 درصد برگ برهان به جای یونجه، پتانسیل تولید گاز در شترهای تغذیه شده با جیره آزمایشی نسبت به جیرهی بدون برهان بیشتر بوده است، همچنین قابلیت هضم مادهی خشک و الیاف نامحلول در شوینده ی خنثی و اسیدی باگاس در دام ها تفاوت معنیداری نداشتند، بنابراین می توان آنرا در جیرهی شتر تککوهانه استفاده کرد.

صحت روشهای مختلف بیزی در ارزیابی ژنومی صفات آستانهای با معماری ژنتیکی متفاوت
چکیده سابقه و هدف: انتخاب ژنومی که نوعی انتخاب به کمک نشانگرهای ژنتیکی میباشد، اثر همه نشانگرهای ژنتیکی پراکنده در سرتاسر ژنوم را بهطور همزمان برآورد میکند. درنتیجه انتخاب ژنومی بهطور بالقوه توانایی توجیه همه واریانس ژنتیکی صفت را دارد. اساس کار در انتخاب ژنومی عدم تعادل پیوستگی بین نشانگر و جایگاه صفات کمی میباشد. با توجه به کمتر مورد توجه قرار گرفتن ارزیابی ژنومی صفات دارای توزیع فنوتیپی گسسته که اکثراً دارای وراثتپذیری پائین هم می باشند، در این تحقیق صحت ارزیابی های ژنومی صفات آستانه ای و صفات پیوسته حیوانات در قالب سناریوهای مختلفی با استفاده از شبیه سازی رایانهای مورد تجزیه و تحلیل قرار گرفت مواد و روشها: در این مطالعه ژنومی حاوی 1000 نشانگر تک نوکلئوتیدی دو آللی با تراکم 1/0 سانتی مورگان بصورت تصادفی روی کروموزومی به طول 100 سانتی مورگان شبیهسازی شد. تعداد QTLهای10، 50 و 100 و سطوح وراثتپذیری 1/0، 2/0 و 3/0 برای صفات آستانهای و پیوسته در نظر گرفته شد. صفات آستانهای با سطوح فنوتیپی دو، چهار، هشت و شانزده و صفات پیوسته بهعنوان شاهد در سه جمعیت 1000، 2000 و 5000 با سه روش آماری بیز A، بیزB و بیز C مورد ارزیابی قرار گرفت. پیاده سازی سناریوهای مورد نظر در این مطالعه با استفاده از بسته های hypred و BGLR در محیط نرم افزار R انجام گرفت. یافته ها: مقادیر صحت ارزشهای اصلاحی ژنومی صفات مختلف در همه سناریوهای مورد بررسی نشان از افزایش همبستگی بین ارزش های اصلاحی برآورد شده با ارزش های اصلاحی واقعی با افزایش تعداد سطوح فنوتیپی در صفات آستانه ای دارد. دامنه تغییرات صحت های بدست آمده در سناریوهای مختلف برای صفت آستانه ای با دو گروه فنوتیپی از همه صفات دیگر بیشتر بوده است. البته با افزایش تعداد گروه بندی ها تفاوت بین صفات با 2 گروه فنوتیپی با صفات با چهار گروه فنوتیپی قابل محسوس تر می باشد. ولی از 4 سطح فنوتیپی هر چه بالاتر می رویم مقادیر بدست آمده نزدیکتر به هم می باشد. صحت های پیشبینی شده در روشهای مورد مطالعه بدلیل تشابه ماهیت محاسباتی به هم نزدیک بوده ولی تغییر صحت ها نسبت به تغییر تعداد QTL ها برای صفات آستانهای و صفات پیوسته در روش آماری بیز C منظم تر مشاهده می شود. با افزایش وراثتپذیری صفت، صحت برآورد اثرات آللی و متعاقب آن پیشبینی ارزشهای اصلاحی ژنومی افزایش یافت. کمترین و بیشترین مقادیر صحت ارزش های اصلاحی ژنومی به ترتیب برای صفات آستانهای دوسطحی آنالیز شده با روش بیز B در سطح وراثتپذیری 1/0 و صفات پیوسته آنالیز شده با روش بیزA در سطح وراثتپذیری 3/0 ثبت شد. برای پیشبینی ارزشهای اصلاحی ژنومی در انتخاب ژنومی نیاز به برآورد اثرات آللی (SNP) در جمعیت مرجع میباشد. لذا افزایش تعداد QTLها از 10 به 100 و افزایش تعداد افراد جمعیت مرجع از 1000 به 5000 فرد منجر به افزایش میزان صحت پیشبینی ارزشهای اصلاحی ژنومی در همه صفات آستانهای و صفات پیوسته گردید. نتیجه گیری: برای ارزیابی ژنومی صفات آستانهای استفاده از روشهای آستانهای را می توان جایگزین روشهای استاندارد کرد. چون ارزیابی ژنومی صفات آستانهای با روشهای استاندارد منجر به کاهش صحت ارزیابیهای ژنومی میگردد، حتی برای صفات با 16 گروه فنوتیپی هم استفاده از روشهای آستانه ای در همه سناریوها منجر به دقت بیشتر ارزشهای اصلاحی ژنومی نسبت به روشهای استاندارد گردید. افزایش گروهبندی صفات آستانهای منجر به افزایش صحت ارزش های اصلاحی ژنومی می گردد، ولی بیش از 8 سطح فنوتیپی برای صفات آستانه ای توصیه نمی شود، چون تأثیر چندانی در افزایش صحت ارزیابیهای ندارد.
