Bovine rotavirus is one of the main causes of diarrhea in calves at the age of one to two weeks. Both vp4 and vp7 proteins form the outer capsid of the virus, producing antibodies and stimulating the immune system. Camel lactoferrampine-Lactoferrin is a potent antiviral that binds to heparin proteoglycine sulfate, prevents virus entry into the host cell and can replace antiviral drugs. The aim of this study was to investigate the possibility of replacement of camel lactoferrin peptide with antiviral drugs.Materials and methods: In this study, the possibility of replacing six recombinant camel lactoferrin peptides named clf36-1, clf36-2, 12A-36F + BC, A_Scaning, AF, Arm with antiviral drugs and efficient binding of these peptides to Surface proteins of bovine rotavirus (vp4 and vp7) was investigated by molecular dynamics simulation using GROMACS software and they were compared due to their bond-dissociation energy in docking step and amount of electric charge before Neutralizing the electric charge of systems in molecular dynamics and finally the best substituted peptide was selected according to Radius of gyration diagram which is the main determinative factor in system efficiency in vivo and also indicates the possibility of stable binding of recombinant lactoferrin peptide to the virus’ surface protein.Results: The results of this study indicate that the lactoferrin peptide binds well with the rotavirus’ surface proteins and can thus prevent the virus from entering the cells. However, among the six peptides examined, the 12A-36F + BC peptide in both vp4 and vp7 systems seems to be inefficient in vivo in spite of the highest bond-dissociation energy and the most positive electric charge before Neutralizing the electric charge of systems in molecular dynamics. However, in vivo experiments are needed to prove the efficacy of this peptide.}, keywords = {Bovine diarrhea,Bovine rotavirus,Camel lactoferrampine-Lactoferrin,molecular dynamics simulation}, title_fa = {بررسی بیوانفورماتیکی میانکنش شش پپتید نوترکیب لاکتوفرین شتری با پروتئین‌های سطحی روتا ویروس گاوی}, abstract_fa = {سابقه و هدف: اسهال یک بیماری رایج میان گوساله ها بوده که سبب زیان اقتصادی چشمگیر در صنعت گاوهای شیری و گوشتی به علت افزایش میزان مرگ و میر، هزینه‌های درمان و کاهش نرخ رشد و افزایش سن اولین زایمان می‌گردد. روتاویروس گاوی یکی از عوامل اصلی ابتلا به اسهال در گوساله ها در سن یک تا دوهفتگی است. دو پروتئین vp4 و vp7 کپسید خارجی ویروس را ساخته و سبب تولید آنتی بادی و تحریک سیستم ایمنی می‌شوند. لاکتوفرامپین-لاکتوفریسین شتری یک ضد ویروس قوی است که با اتصال به هپارین سولفات پروتئوگلایسین مانع ورود ویروس به سلول میزبان شده و می‌تواند جایگزین داروهای ضد ویروسی شود. هدف از این پژوهش بررسی امکان جایگزینی پپتید لاکتوفرین شتری با داروهای ضد ویروسی می باشد.مواد و روش ها: در این مطالعه امکان جایگزینی شش پپتید نوترکیب لاکتوفرین شتری به نام‌های clf36-1، clf36-2، 12A-36F+BC، A_Scaning، AF، Arm با داروهای ضدویروسی و اتصال کارآمد این پپتیدها با پروتئین‌های سطحی روتاویروس گاوی(vp4 و vp7) با استفاده از روش شبیه‌سازی دینامیک مولکولی و با استفاده از نرم افزار گرومکس بررسی و با توجه به انرژی آزاد پیوند شدن لیگاند-پروتئین در مرحله داکینگ و میزان بار الکتریکی سیستم پیش از خنثی سازی بار در دینامیک مولکولی مورد مقایسه قرار گرفته و در نهایت بهترین پپتید جایگزین با توجه به نمودارهای شعاع چرخشی که مهم ترین عامل تعیین کننده در کارآمدی سیستم‌ها در شرایط درون تنی و همچنین نشان دهنده‌ی امکان اتصال پایدار پپتید نوترکیب لاکتوفرین با پروتئین سطحی ویروس است؛ از میان شش پپتید مورد مطالعه، انتخاب گردید. یافته ها: نتایج حاصل از این پژوهش نشان می دهد پپتید لاکتوفرین به خوبی با پروتئین های سطحی روتاویروس اتصال برقرار کرده و به این وسیله می تواند مانع ورود ویروس به درون سلول‌ها گردد. با این حال از میان شش پپتید مورد بررسی، پپتید 12A-36F+BC در دو سیستم vp4 و vp7 علی رغم داشتن بیشترین انرژی آزاد پیوند شدن لیگاند-پروتئین در بین مدل‌های مورد بررسی در مرحله داکینگ و مثبت‌ترین بار الکتریکی پیش از خنثی سازی بار در دینامیک مولکولی، به دلیل unfold بودن و تغییرات زیاد Rg آنها در طول زمانِ شبیه سازی به نظر می رسد در شرایط بدن اتصال کارآمدی نداشته باشند؛ از این رو با توجه به نمودارهای شعاع چرخشی از مطالعه حاضر انتظار می‌رود سیستم پروتئین سطحی vp4 و پپتید نوترکیب clf36-1 دارای بهترین اتصال باشد.نتیجه گیری: نتایج حاصل از این تحقیق نشان می‌دهد پپتید clf36-1 از میان شش پپتید مورد بررسی طی زمان شبیه سازی پایدار و دارای اتصال کارآمد و مناسب با پروتئین سطحی vp4 بوده، ضمن اینکه طی زمان شبیه سازی ساختار خود را حفظ کرده و از این طریق میتواند مانع ورود ویروس به درون سلول‌ها شده و جایگزین مناسبی برای داروهای ضد ویروسی باشد. با این حال انجام آزمایشات در شرایط in vivo به منظور اثبات اثربخشی این پپتید ضروری می نماید.}, keywords_fa = {اسهال گوساله ها,روتاویروس گاوی,لاکتوفرامپین-لاکتوفریسین شتری,شبیه سازی دینامیک مولکولی}, url = {https://ejrr.gau.ac.ir/article_5044.html}, eprint = {https://ejrr.gau.ac.ir/article_5044_d6ae153de904962e0bae991c93fb03c9.pdf} }
