According to researches, the activity of rumen enzymes also varies with regard to the rumen status, especially pH, and the inherent nature of buffers in improving the rumen's environmental conditions, as well as the possible effect of the physical form of the concentrate on ruminal activity. The present study aimed to investigate the effect of the physical form of the concentrate (pellet and mash) and buffer type of ration (bicarbonate sodium and sesquisodium carbonate) on the activity of some ruminal hydrolytic enzymes including carboxymethyl cellulase, microcrystalline cellulase, Filter paper decomposition activity, protease and alpha amylase in different segments of rumen fluid (including solid, extracellular and intracellular sections) and nitrogen retention and blood hematology of Dalagh fattening lambs. Materials and Methods: In order to investigate the effect of the physical form of the concentrate and the buffer type of diet on the activity of some hydrolytic ruminal enzymes, nitrogen retention and blood hematology in Dalagh fattening lambs, used 28 male lambs in 6±1.2 months and average weight of 28±2.7. This experiment was conducted in a completely randomized design with a 2 × 2 factorial arrangement and a 98-day period (14 days of adaptation and 84 days of the main period) with 4 treatments and 7 replications. Rumen fluid sampling was performed to measure the activity of some hydrolytic enzymes during the final week of the experiment. Also, on day 60, 4 lambs were randomly selected from each treatment and transferred to metabolic cages. After 3 days of adaptation, for 6 consecutive days, the urine and faces were collected so that the nitrogen balance was also measured using nitrogen consumption and excretion It was estimated through urine and faces. To determine the blood parameters, in final day, 3 hours after the morning feeding, blood was taken from the vein and immediately transferred to the laboratory. Blood parameters such as white blood cells, neutrophils, eosinophil’s, lymphocytes and monocytes were measured and red blood cells, hemoglobin, hematocrit, mean blood cell counts, mean hemoglobin, and mean hemoglobin concentration were also determined. Results: Based on the results, change in physical form of the concentrate and the buffer type did not have a significant effect on the activity of the total carboxymethyl cellulase enzymes, microcrystalline cellulase, and activity of filter paper decomposition, alpha amylase and protease. Although the experimental diets have been effective in the activity of various segments protease enzymes and filter paper decomposition activity. This effect has not been sufficient to alter the overall activity of these enzymes. In relation to nitrogen balance, no significant difference was found between nitrogen intake, total nitrogen and nitrogen retention in experimental groups, and only excreted nitrogen through feces under the influence of sesquisodium carbonate buffer increased compared to those receiving sodium bicarbonate buffer. Also, no significant differences were found between the hematological parameters of the lambs. Conclusion: This study showed that the physical form of the concentrate and the buffer type had a significant difference in the activity of the total carboxymethyl cellulase enzymes, microcrystalline cellulase, activity of filter paper decomposition, alpha amylase and protease and nitrogen balance, as well as there are no, between hematology parameters.}, keywords = {Nitrogen retention,Buffer,Physical form of concentrate,enzyme activity of rumen,hematology}, title_fa = {تاثیر شکل فیزیکی کنسانتره و نوع بافر جیره بر فعالیت برخی از آنزیم های هیدرولیتیک بخش های مختلف شیرابه شکمبه، ابقای نیتروژن و هماتولوژی در بره های پرواری دالاق}, abstract_fa = {چکیده سابقه و هدف: در پرواربندی گوسفندان، بدلیل مصرف جیره ای که حاوی میزان کنسانتره بالا و علوفه پایین ، بر هم خوردن تعادل محیط شکمبه امری یحتمل به نظر می‌رسد. باتوجه به تحقیقات صورت گرفته فعالیّت آنزیمهای شکمبه نیز با توجه به وضعیت شکمبه بخصوصpH تغییر میکند و خاصیت ذاتی بافر ها مبنی بر بهبود شرایط محیطی شکمبه و همچنین تاثیر احتمالی شکل فیزیکی کنسانتره بر فعالیت شکمبه، تحقیق حاضر جهت بررسی تاثیر شکل فیزیکی کنسانتره(پلت و آردی) و نوع بافر جیره (بی‌کربنات‌سدیم و سسکویی‌سدیم‌بی‌کربنات) بر فعالیّت برخی آنزیمهای هیدرولیتیک شکمبه شامل کربوکسی متیل سلولاز، میکروکریستالین سلولاز، فعالیّت تجزیه کاغذ صافی، پروتئاز و آلفا آمیلاز در بخشهای مختلف شیرابه شکمبه (شامل بخش جامد، خارج سلولی و درون سلولی) و ابقای نیتروژن و همچنین هماتولوژی خون بره های پرواری دالاق انجام گرفت. موادوروش ها: به منظور بررسی تاثیر شکل فیزیکی کنسانتره و نوع بافر جیره بر بر فعالیّت برخی آنزیم‌های هیدرولیتیک شکمبه، ابقای نیتروژن و هماتولوژی خون در بره های پرواری نژاد دالاق از 28 رأس بره ی نر 2/1±6 ماهه با میانگین وزن 7/2±28 استفاده شد.این آزمایش در قالب طرح کاملا تصادفی به صورت فاکتوریل2*2 و در دوره 98 روزه ( 14روز عادت پذیری و 84 روز دوره اصلی) با 4 تیمار و7 تکرار انجام شد. نمونه گیری از مایع شکمبه جهت اندازه گیری فعالیّت برخی آنزیم های هیدرولیتیک شکمبه در هفته ی پایانی دوره اصلی صورت گرفت تا آزمایش های مربوط به این پارامتر انجام شود. همچنین در روز 60 پرواربندی 4رأس بره از هر تیمار بطور تصادفی انتخاب و به قفس های متابولیکی منتقل شدند وبعد از 3روز عادت پذیری به مدت 6 روز متوالی، ادرار و مدفوع گوسفندان جمع آوری شد تا تعادل نیتروژن نیز با استفاده از مقادیر مصرف و دفع ازت از طریق ادرار و مدفوع برآورد شد. برای تعیین فراسنجه هـای خـونی روز پایانی 3 ساعت بعد از وعده خوراک مصرفی صـبح، خون گیری از سیاهرگ وداج گرفته شد و بلافاصـله به آزمایشگاه انتقال یافـت. پارامترهای خونی از جمله گلبولهای سـفید خون، نوتروفیل، ائوزینوفیل، لمفوسیت و مونوسیت اندازه گیـری شـد و گلبولهـای قرمـز خـون، هموگلـوبین، هماتوکریـت، حجـم متوسـط گلبولهای قرمـز، میـانگین هموگلـوبین سـلولی و میـانگین غلظـت هموگلوبین سلولی نیز تعیین گردید. یافته‌ها: براساس نتایج به دست آمده تغییر شکل فیزیکی کنسانتره و نوع بافر تاثیر معنی‌داری در فعالیت کل آنزیم های کربوکسی متیل سلولاز، میکروکریستالین سلولاز(آویسلاز)، فعالیّت تجزیه کاغذ صافی، آلفا آمیلاز و پروتئاز ندارد. با وجود این که جیره تیمارهای آزمایشی در فعالیت بخش‌های مختلف آنزیم پروتئاز و فعالیّت تجزیه کاغذ صافی اثر گذار بوده است اما این تاثیر به حدی نبوده که فعالیت کل این آنزیم ها را تغییردهد. در رابطه با توازن نیتروژن نیز اختلاف معنی داری بین نیتروژن مصرفی، مجموع نیتروژن دفعی و ابقا نیتروژن در بین گروه های آزمایشی یافت نشد و تنها نیتروژن دفعی از طریق مدفوع تحت تاثیر بافر سسکویی سدیم کربنات نسبت به گروه های دریافت کننده بافر بی کربنات سدیم افزایش یافت. همچنین هیچ گونه اختلاف معنی داری بین پارامترهای هماتولوژی بره ها نیز مشاهده نشد. نتیجه‌گیری: این مطالعه مشخص نمود که شکل فیزیکی کنسانتره و نوع بافر اختلاف معنی داری را در فعالیت کل آنزیم های کربوکسی متیل سلولاز، میکروکریستالین سلولاز(آویسلاز)، فعالیّت تجزیه کاغذ صافی، آلفا آمیلاز و پروتئاز و توازن نیتروژن و همچنین داری بین پارامترهای هماتولوژی بره ها ندارد.}, keywords_fa = {ابقای نیتروژن,بافر,شکل فیزیکی کنسانتره,فعالیت آنزیمی شکمبه,هماتولوژی}, url = {https://ejrr.gau.ac.ir/article_4215.html}, eprint = {https://ejrr.gau.ac.ir/article_4215_4f42931fa1ccf358a89c0908c2ad9477.pdf} }
